The city occupies the northwestern shore of Lake Ontario within the Regional Municipality of Peel, bordered by Toronto's Etobicoke district to the east, Brampton to the north, Milton to the northwest, and Oakville to the southwest. After Toronto itself, it is the second-largest municipality in the Greater Toronto Area.

The city's commercial character is shaped by three intersecting forces. Mississauga's civic anchor set is unusually strong for a suburban-regional market. Two of the five clusters attach to Mississauga Hospital and one attaches to Credit Valley Hospital. Both hospitals are operated by Trillium Health Partners, the regional acute-care network for the western Greater Toronto Area, and together they form one of the largest community hospital systems in Ontario. A fifth cluster includes a LifeLabs Medical Laboratory Services site, extending the medical footprint into the outpatient diagnostic layer.

The academic anchor is the University of Toronto Mississauga, a full research campus of the University of Toronto with an enrolment in the range of fifteen thousand students.
